You use "may" when asking permission-- "can" usually refers to ability. Thus, if someone asks "Can I have a glass of water?" a smartass might reply, "Yes you may."

When and how to correctly use these two words also depends on whether you are asking or answering a question in which "may" or "can" is used. When asking a question, it is considered more polite and formal (For example: "May I have a glass of water?" as opposed to "Can I have a glass of water?"). The same applies for when you are answering someone. Also, "may" is usually used when you are asking or allowing something.
