Is there a difference between 저는 and 저의?

저의 가방입니다. It's my bag.

‘저는’ is ‘as me’ or ‘for me’
‘저의’ is ‘my’ or ‘of me’

First, '저' is 'formal I'.
 

"는" has many meanings but I think these two are fine.

1. Contrast with others.
ex) 너는 할 수 있지만 나는 할 수 없다; 'You' can do it, but 'I' can't do it.
2. Showing the topic of the sentence.
ex) 너는 점심을 먹는다; 'You' are having lunch.
 

"의" is postposition for making word possessive.

When it attached to a certain word, its meaning will change to 'Possessive'.
So, "저의" means "My".

Yes, there is a difference. In simple terms:

저는 (jeo-neun): ”I am” or ”I” (with a topic marker)
저의 (jeo-ui): ”my” or ”mine” (possessive)

’저’ is used when ’I’ lower myself.
’저는’ is ’I’ as the topic, as in ’저는 걸을 것입니다.’ (I will walk.)
’저의’ is ’my’, as in ’저의 상자는 가볍습니다.’ (My box is light.)
Note that one may use ’제’ instead of ’저의’.

And 저의 means ”my”
